The present study was carried out to investigate toxico-pathological effect of dexamethasone in Wistar rats. Out of four groups of rats (6 rats/sex/group), one group was kept as control and other three groups were treated with dexamethasone alone at three different dose levels (2.5, 6.25 and 12.5 mg/kg). Dexamethasone formulated in filtered water was administered by oral route to the treatment group of animals for a period of two weeks and control group animals received vehicle alone. At the end of the treatment period, all surviving rats were subjected to terminal necropsy. Absolute organ weight of adrenals, heart, kidneys, liver, spleen, testes, ovaries, epididymides, uterus, seminal vesicles, thymus, lungs and brain were taken to estimate relative organ weight. Organs showing gross pathological findings were processed and histopathological examination was performed. Two weeks repeated administration of dexamethasone revealed significant decrease in absolute weight of adrenals, spleen and thymus in both the sexes. Similar findings were evident in relative organ weight of both male and female rats from treatment groups as compared to control group. Histopathological findings in treated groups revealed atrophy of adrenal cortex, fatty infiltration of femur bone and decreased bone marrow mass, fatty infiltration and congestion in liver, alveolar histiocytosis in lungs, lymphoid depletion in spleen and lymph nodes.The present study was carried out to investigate toxico-pathological effect of dexamethasone in Wistar rats.
